Inducible Knockout Mouse

Learn how to generate inducible knockouts in mice.

 

Knock outs which are embryonic lethals or toxic (kill the animal eventually) can be made inducible. This has the advantage of allowing the knockout to take place AFTER development / embryogenesis has occurred. This is accomplished by using a Tet On / Tet Off system. Using the tet system, the inducer can be added to the drinking water of the animal when you want to start the knock out. After drinking the water for several days, complete knockout of the gene can occur. This is done by expressing CRE recombinase with the inducer.

Insertion of an IRES in between two open reading frames ( gene to knock out – IRES – Reporter gene) allow one to use LacZ or GFP reporter genes to visualize the area which is knocked out in the mouse.
